Molecular inhibitors of DNA repair: searching for the ultimate tumor killing weapon

In our recent publication which is a collaboration of our laboratory in NTUA (Athens, Greece) with Dr. Michalopoulos' bioinformatics lab in Biomedical Research Foundation of the Academy of Athens (BRFAA-Athens), we target the critical pathways of DNA repair. We perform an analysis of the main repair pathways responsible for processing complex DNA damage. At the same time we have included all current NCI clinical trials on DNA repair inhibitors. By applying bioinformatic tools, we identify repair proteins implicated in more than two phenomenically distinct DNA repair pathways (e.g., base excision repair and nonhomologous end joining), that is, the concept of ‘synthetic lethality.’ Our study can aid towards the optimization of this therapeutic strategy and, therefore, maximizing treatment effectiveness like in the case of radiation therapy.
